动物体内RNA转染试剂18668-11-1

Entranster^TM-in vivo是英格恩生物公司（Engreen Biosystem Co,Ltd.）最新研发合成的用于动物体内转染的转染试剂。Entranster^TM-in vivo动物体内转染试剂可将小片断RNA如siRNA，miRNA，mimic、inhibitor转染到动物体内，在动物体内进行RNA干扰等实验。

Entranster^TM-in vivo试剂不是病毒，改变了病毒转染方法复杂、周期长、不稳定、花费大、安全性等诸多问题。Entranster^TM-in vivo转染效率高，方法简单，无需病毒包装，直接将RNA送入目的细胞的细胞核中表达。

Entranster^TM-in vivo，非脂质体，采用纳米技术合成，通过保护和浓缩核酸，避免核酸被体液破坏，并通过物理作用将核酸富集在细胞表面，增强核酸进入细胞组织，从而产生作用。Entranster^TM-in vivo能耐受体内复杂的环境，不产生免疫反应。

方法简单

如图1所示，只需要将核酸和转染试剂分别稀释，混合，无需过滤，即可直接注射动物。 转染效果 将Entranster^TM-in vivo与Ambion in vivo siRNA(作用于凝血因子VII)和阴性siRNA通过尾静脉注射成年小鼠，2天后，取动物肝脏检测。在mRNA水平和蛋白水平观察干扰效果。见上图，图中最左侧组为对照组注射阴性siRNA为3mg/kg，后边3组为每kg动物注射阳性siRNA量分别为1mg/kg,2mg/kg和3mg/kg情况。

各器官转染 根据推荐用量注射Entranster^TM-in vivo和siRNA（作用于Lamin A/C）和阴性siRNA到成年小鼠。注射后2天收集相应的组织，分离RNA，用qRT-PCR分析Lamin A/C基因的表达水平。图4为尾静脉结果，图5为各器官分别局部注射结果。
